Whole Cell Patch-Clamp and SH-SY5Y tumor cells

چکیده
Human neurobalstoma cells, SH-SY5Y, are widely studied and data on their electrophysiological characteristics can be found in literature. We used this cell line to set up the whole cell patch-clamp technique, not yet available in our institute, with the goal to make it suitable both for our future scientific projects and for interdisciplinary studies with other research groups. Some instruments, being outdated, have been repaired and adapted to obtain the best possible results; others were kindly borrowed by colleagues. Whole cell patch-clamp recordings were obtained and they were highly comparable with data found in literature confirming both these data and the goodness of our setup, although it needs to be updated to most recent scientific standards.

Functional changes in sodium conductances in the human neuroblastoma cell line SH-SY5Y during in vitro differentiation.
1. The electrophysiological properties of voltage-dependent sodium currents were studied in the human neuroblastoma cell line SH-SY5Y before and after in vitro differentiation with retinoic acid, with the use of the whole cell variant of the patch-clamp technique. 2. Voltage steps from a holding level of -90 mV to depolarizing potentials elicited, in both undifferentiated and differentiated cel...

Acid-sensing ion channels (ASICs) have been reported to play a role in the neuronal dopamine pathway, but the exact role in neurotransmitter release remains elusive. Human neuroblastoma SH-SY5Y is a dopaminergic neuronal cell line, which can release monoamine neurotransmitters. In this study, the expression of ASICs was identified in SH-SY5Y cells to further explore the role of ASICs in vesicul...
متن کاملThe large conductance Ca2+ -activated K+ (BKCa) channel regulates cell proliferation in SH-SY5Y neuroblastoma cells by activating the staurosporine-sensitive protein kinases
Here we investigated on the role of the calcium activated K(+)-channels(BKCa) on the regulation of the neuronal viability. Recordings of the K(+)-channel current were performed using patch-clamp technique in human neuroblastoma cells (SH-SY5Y) in parallel with measurements of the cell viability in the absence or presence of the BKCa channel blockers iberiotoxin(IbTX) and tetraethylammonium (TEA...
متن کاملThe neuroprotective effect of BSA-based nanocurcumin against 6-OHDA-induced cell death in SH-SY5Y cells
Objective: Parkinson’s disease (PD) is regarded as the second most common neurodegenerative disease affecting elderly population. There is a tendency toward finding natural cures to suppress the initiation and progression of this disease. Some epidemiological studies indicated lower incidence of PD in populations that consume curry. Curcumin, as the main ingredient of turmeric, has been suppose...
متن کاملStereospecific interaction of ketamine with nicotinic acetylcholine receptors in human sympathetic ganglion-like SH-SY5Y cells.
BACKGROUND Surprising clinical evidence suggests a block of sympathetic transmission by ketamine. The action of ketamine on nicotinic acetylcholine receptors (nAChRs) in human ganglions is unknown. Because ganglionic transmission depends on nAChRs, such information may help to clarify whether ketamine impairs ganglionic transmission in men. Because racemic ketamine as well as S(+)-ketamine are ...
